Medicinal mushrooms have been used in human medicine for thousands of years. Here's what the evidence actually says about the most studied mushrooms for dogs.
How Medicinal Mushrooms Work
The primary active compounds in medicinal mushrooms are beta-glucans — complex polysaccharides found in the cell walls of the mushroom. Beta-glucans are biological response modifiers: they don't directly attack pathogens or tumors, but they modulate the immune system's ability to recognize and respond to them.

One critical note on sourcing: the active compounds are concentrated in the fruiting body (the actual mushroom) not the mycelium (the root structure). Turkey Tail (Trametes versicolor)

Turkey tail has the strongest clinical evidence of any medicinal mushroom in dogs. A randomized, placebo-controlled clinical trial at the University of Pennsylvania School of Veterinary Medicine gave turkey tail extract to dogs with hemangiosarcoma — one of the most aggressive cancers in dogs. The active compounds — PSK (polysaccharide-K) and PSP (polysaccharide-peptide) — are among the most studied anti-cancer compounds in both human and veterinary oncology. PSK is an approved cancer treatment drug in Japan.
Turkey tail also supports gut microbiome diversity. A human clinical trial found it significantly increased populations of beneficial Bifidobacterium and Lactobacillus while reducing pathogenic bacteria — relevant for dogs with chronic digestive issues.
